Pol I Inhibitor (CX-3773)

The second generation of RBIs created from Cylene’s proprietary RABIT™ technology platform represents a novel class of fluoroquinolones with ADMET/PK properties different from that of CX-3543. CX-3773 targets the Pol I transcription complex and has unique PK and biodistribution properties suitable for use against primary brain tumors and brain metastases. Furthermore, CX-3773 has a lower molecular weight than CX-3543, and readily crosses the blood-brain-barrier, distributing widely into the brain tissue. It therefore has the potential to be a novel compound for the treatment of primary brain tumors and brain metastases. Essentially no metabolism can be detected from this compound.

Moreover, CX-3773 demonstrates long duration of action, making it an attractive agent to use as a single agent or in combination with other anti-tumor drugs. This is an important and differentiating feature, in that most anti-tumor compounds can not cross the blood-brain barrier and therefore can not be used to treat brain tumors. Treatment of brain tumors and brain metastases represents a significant unmet medical need.
